We developed peanut allergen-specific aptamers using in silico computational methods. These aptamers specifically bind to peanut allergens Ara h2 and Ara h1. Our aptamers effectively nautralize crude peanut-induced degranulation in the RBL-2H3 cell line in vitro. Additionally, we found that our aptamers are stable at various temperatures, demonstrating excellent heat stability.

To further test their application, we introduced these aptamers to peanut plants by watering them with an aptamer solution.
We found that the aptamers were only present in the peanut seeds.
Remarkably, peanuts grown this way did not induce degranulation in the RBL-2H3 cells, indicating that we successfully developed allergy-free peanuts.
Our aptamers also proved beneficial in food processing applications. When mixed with commercial peanut butter, they effectively neutralized the peanut allergens by stably binding to denatured allergenic peanut proteins. Additionally, coating or soaking peanut seeds with these aptamers neutralized the allergenic proteins, preventing degranulation.
We verified that our aptamers are non-immunogenic and can modulate the Th1/Th2 immune response in allergy models. Specifically, they induce Th1 responses while downregulating Th2 responses, which are key markers in the development of allergies. Our aptamers effectively downregulate Th2 cytokines IL-4, IL-5, and IL-13, while inducing Th1 cytokines. Furthermore, we demonstrated that these aptamers are effective both preventatively and therapeutically.
